(2) reported high proportions of MM-i in serum in improving or stablemuscle disease and reduced proportions in deteriorating disease. However, Annesley et al. used the reverse nomenclature, whereby MM-i refers to the most cathodal MM sub-band. Misinterpretation of reported observationsbecause ofnomenclatureconfusionhighlights the need for an accepted nomenclature. We would favor the immediate adoption of the convention whereby MM-i represents the most anodal sub-band. Where the MM isoenzyme is designed as CK-3, the subtypes should begiven lower case letters (3a, 3b, 3c) consecutively from the anode. A similar convention should apply to the naming ofMB (OK-2) sub-types.

Protein analysis with bicinchoninic acid.

Some of the variables and the interferences which affect the reaction kinetics of proteins with bicinchoninic acid (BCA) (4,4'-dicarboxy-2,2'-biquinoline) have been studied. Interferences attributed to glucose, ascorbic acid, and uric acid in this reaction were significantly reduced by performing a two-point assay and by including borate ions in the buffer. The method was shown to be suitable f...

Development and characterization of the NanoOrange protein quantitation assay: a fluorescence-based assay of proteins in solution.

We developed a sensitive fluorescence assay for the quantitation of proteins in solution using the NanoOrange reagent, a merocyanine dye that produces a large increase in fluorescence quantum yield upon interaction with detergent-coated proteins. The NanoOrange assay allowed for the detection of 10 ng/mL to 10 micrograms/mL protein with a standard fluorometer, offering a broad, dynamic quantita...

Microwave-assisted method for microassay of proteins using bicinchoninic acid.

A microwave-assisted microassay has been developed for estimation of proteins, using bicinchoninic acid (BCA). The protocol described here is rapid with a reaction time of merely 95 sec, requires lesser amounts of BCA and other reagents, and tolerates larger or comparable concentration of various interfering substances.

Colorimetric protein assay techniques.

There has been an increase in the number of colorimetric assay techniques for the determination of protein concentration over the past 20 years. This has resulted in a perceived increase in sensitivity and accuracy with the advent of new techniques. The present review considers these advances with emphasis on the potential use of such technologies in the assay of biopharmaceuticals. The techniq...

Quantitative evaluation of proteins with bicinchoninic acid (BCA): resonance Raman and surface-enhanced resonance Raman scattering-based methods.

A rapid and highly sensitive bicinchoninic acid (BCA) reagent-based protein quantitation tool was developed using competitive resonance Raman (RR) and surface-enhanced resonance Raman scattering (SERRS) methods. A chelation reaction between BCA and Cu(+), which is reduced by protein in an alkaline environment, is exploited to create a BCA-Cu(+) complex that has strong RR and SERRS activities. U...
